ok, so I put an application in on Monday at a place I really want to get a job at...she said she would look to see if anything was available, and call me back.
well, she didnt call be on Tuesday, so I called her wednesday, but she was at a meeting, so I left her a message on her voice mail asking if she reviewed my application and blah blah blah and to call me back.
she didnt call back.
do you think I should call again today? or wait til tomorrow?

tough call... you dont want to be overly pushy or annoying to them... but its rude of her not to call you back at all... even to let you know nothing is available...
i would say wait until tomorrow morning... give her today to get back to you

is this for teaching?
if its anything similar to ny, i would just wait for them to call you...
i hate to be discouraging, but i put in literally around 100 applications, and received in all, interviews with 7 different schools..from those 7 i still wasnt hired for a full time job until 11 months after i put in my first application

no, its not for teaching...its to be a TSS, part time in the evenings while im subsitute teaching

Ditto. I'd wait til Monday. I know you want to know and you want to show initiative, but it's a fine line between initiative and being pushy.
