I have to get from LI to Madison Avenue between 59th street and 60th street. What is the best route?

If youre taking the train, Id say the N, W, or R train to Lexington.

honestly hop in a cab outiside penn...
you can take the 4/5 but you would have to take the 2/3 to 42nd, xfer to the shuttle to grand central and then pick up the 4/5 there.
Or you can just take the 2/3 uptown but then have to walk from 7th ave to madison which is kinda long.

I used to work around there and when I got off at Lex I only had to walk like 3-4 blocks.

From Penn? I would walk the one black to the N/R - follow Macy's and you can't miss it, and then take that to the 59th street and Lex stop - then walk two blocks west to Madison.
Or you can take the E train to 51st and Lex and walk from there, or transfer to the 4/5/6 to 59th.

THE N/W/R goes to 34th Street which is one block from penn. When you get out, literally look for Macy's and walk along the length of it - the whole block - and you'll run into the subway.

directly from penn you can take the E to Lex Avenue and then take the six train one stop to 59th and Lex. it's pretty easy. OR do what the others said and walk to the N/R/W - they all stop at 59th street.

You can take the E train right inside Penn to 5th Ave/53rd. Come out on the Madison Ave side and walk straight up the 6 blocks.

if you don't mind walking you can take the E to 5th and 53rd and walk to 59th

You can take the LIRR to Penn then take the F,E,V ,N,W,R if Im not mistaken can be found around Penn st, to 59th St and just walk a bit
